Caffeic acid improves cell viability and protects against DNA damage: involvement of reactive oxygen species and extracellular signal-regulated kinase BJMBR
Li,Y.; Chen,L.J.; Jiang,F.; Yang,Y.; Wang,X.X.; Zhang,Z.; Li,Z.; Li,L..
Hormesis is an adaptive response to a variety of oxidative stresses that renders cells resistant to harmful doses of stressing agents. Caffeic acid (CaA) is an important antioxidant that has protective effects against DNA damage caused by reactive oxygen species (ROS). However, whether CaA-induced protection is a hormetic effect remains unknown, as is the molecular mechanism that is involved. We found that a low concentration (10 μM) of CaA increased human liver L-02 cell viability, attenuated hydrogen peroxide (H2O2)-mediated decreases in cell viability, and decreased the extent of H2O2-induced DNA double-strand breaks (DSBs). In L-02 cells exposed to H2O2, CaA treatment reduced ROS levels, which might have played a protective role. CaA also activated the...

Changes in growth, pigment content and antioxidants in the root and leaf tissues of wheat plants under the influence of exogenous salicylic acid Braz. J. Plant Physiol.
Sahu,Gopal Krishna; Sabat,Surendra Chandra.
The effect of various concentrations of salicylic acid (SA) on the growth, pigment content and the activity of antioxidants was investigated in the laboratory grown wheat plants. The root and shoot growth was affected at higher concentration of SA in early days of growth. The activities of catalase (CAT), ascorbate peroxidase (APX) and guaicol-specific peroxidase (POX) declined with the application of SA (50, 500 and 1000 µM), the decrease being more pronounced with the increase in SA concentrations both in the root and leaf tissues. On the other hand superoxide dismutase (SOD) activity increased with the application of SA. At low concentrations, SA has no effect on the activities of these enzymes in vitro. Salicylic acid at higher concentrations (5- and...

Changes in nitrate reductase activity and oxidative stress response in the moss Polytrichum commune subjected to chromium, copper and zinc phytotoxicity Braz. J. Plant Physiol.
Panda,Sanjib Kumar; Choudhury,Shuvasish.
The main aim of this paper was to investigate the effect of chromium (Cr), copper (Cu) and zinc (Zn) on nitrate reductase (NR) activity and oxidative stress responses in the moss Polytrichum commune. Cr, Cu and Zn resulted in the inhibition of NR activity. A decline in total chlorophyll content was observed after 24 and 48 h of metal treatment. Accumulation of the metals showed a dose and time dependent increase. High accumulation of Cu, Cr and Zn were seen in moss shoots after 24 and 48 h of treatment. Treatment of Cr, Cu and Zn for 24 or 48 h resulted in the increase of malondialdehyde (MDA) content in moss shoots. The highest increase was observed in shoots under Cu treatment followed by Cr and Zn. The MDA content was significantly higher after 48h....

Characterization of Seed Oils of Pterodon emarginatus Vogel (Sucupira) Obtained by Different Extraction Methods BABT
Costa-Singh,Tainara; Jorge,Neuza.
Abstract The present study aims to evaluate the influence of extraction methods on the quality of bioactive compounds and antioxidant capacity in Pterodon emarginatusVogel (sucupira) oils. The oils were extracted from the sucupira seeds by Soxhlet and by the extraction system of Bligh & Dyer. The oils were analyzed as to fatty acid profile, tocopherols, phytosterols, carotenoids, total phenolics, and total antioxidant capacity. The extraction by Soxhlet showed better yield of total lipids and was more efficient to extract tocopherols, phytosterols, and carotenoids, besides presenting better antioxidant activity by DPPH•. However, this method showed insufficient capacity to extract the polar lipid components of the sample, as evidenced by the low...

Chromium stress in plants Braz. J. Plant Physiol.
Panda,S.K.; Choudhury,S..
The article presents an overview of the mechanism of chromium stress in plants. Chromium is known to be a toxic metal that can cause severe damage to plants and animals. Chromium-induced oxidative stress involves induction of lipid peroxidation in plants that causes severe damage to cell membranes. Oxidative stress induced by chromium initiates the degradation of photosynthetic pigments causing decline in growth. High chromium concentration can disturb the chloroplast ultrastructure thereby disturbing the photosynthetic process. Like copper and iron, chromium is also a redox metal and its redox behaviour exceeds that of other metals like Co, Fe, Zn, Ni, etc. The redox behaviour can thus be attributed to the direct involvement of chromium in inducing...

Farias,Carine Coneglian de; Bonifácio,Kamila Landucci; Matsumoto,Andressa Keiko; Higachi,Luciana; Casagrande,Rúbia; Moreira,Estefânia Gastaldello; Barbosa,Décio Sabbatini.
Parkinson's disease (PD) is characterized by progressive degeneration of dopaminergic neurons in the substantia nigra pars compacta. Furthermore, oxidative stress plays a role in PD, causing or contributing to the neurodegenerative process. Currently PD has only symptomatic treatment and still nothing can be done to stop the degenerative process of the disease. This study aimed to comparatively evaluate the antioxidant capacity of pramipexole, selegeline and amantadine in different in vitrostudies and to offer possible explanations on the molecular antioxidant mechanisms of these drugs. In vitro, the antioxidant capacity of the drugs was assessed by the ability of antiparkinsonian drugs to decrease or scavenge ROS in the neutrophil respiratory burst,...

Cyclic nitroxides inhibit the toxicity of nitric oxide-derived oxidants: mechanisms and implications Anais da ABC (AABC)
Augusto,Ohara; Trindade,Daniel F.; Linares,Edlaine; Vaz,Sandra M..
The substantial therapeutic potential of tempol (4-hydroxy-2,2,6,6-tetramethyl-1-piperidinyloxy) and related cyclic nitroxides as antioxidants has stimulated innumerous studies of their reactions with reactive oxygen species. In comparison, reactions of nitroxides with nitric oxide-derived oxidants have been less frequently investigated. Nevertheless, this is relevant because tempol has also been shown to protect animals from injuries associated with inflammatory conditions, which are characterized by the increased production of nitric oxide and its derived oxidants. Here, we review recent studies addressing the mechanisms by which cyclic nitroxides attenuate the toxicity of nitric oxidederived oxidants. As an example, we present data showing that tempol...

Effect of dried extract of boldo (Peumus Boldus Mol.) on growth and oxidative tissue status of broiler chickens Rev. Bras. Ciênc. Avic.
Fellenberg,MA; Delporte,C; Backhouse,N; Peña,I; Speisky,H.
The addition of antioxidants to broiler diets has been shown to enhance their antioxidant status. Since boldo (Peumus boldus Mol.) leaves contain highly antioxidant molecules, a dried extract of boldo (DEB) was added to broiler diets to improve "in vivo" antioxidant tissue status and to favor animal growth. A DEB standardized for antioxidant content was prepared and added to poultry diets at three different levels (low-DELB, medium-DEMB, and high-DEHB) for a period of 6 weeks. A single negative control (no added antioxidant) and one positive control (supplementation with 200 mg/kg vitamin E) were used. Plasma antioxidant capacity (PAC), thiol content (GHS), and basal and induced lipoperoxidation of liver, leg and breast tissues were determined in birds at...

Effect of Harvest Time and L-Cysteine as an Antioxidant on Flesh Browning of Fresh-Cut Cherimoya (Annona cherimola Mill.) Chilean J. Agric. Res.
Campos-Vargas,Reinaldo; Defilippi,Bruno G; Romero Q,Paula; Valdés G,Héctor; Robledo M,Paula; Prieto E,Humberto.
Browning development is the most important factor limiting the quality of fresh-cut cherimoya (Annona cherimola Mill.). However, there is little information available about its causes and methods of control. The effectiveness of L-cysteine (0.125, 0.25 and 0.5%) in fresh-cut cherimoya harvested on two occasions (October and November) and stored for 6 and 12 days at 0 °C was studied. In order to understand the biological basis of browning, polyphenol oxidase (PPO) enzyme activity and total phenolic content in fresh-cut pieces were measured. Quality measurements and sensory analysis indicated that 0.5% L-cysteine was somewhat effective in reducing browning development, without affecting other quality attributes. In terms of physiological parameters, PPO...

Jordao Júnior,Alceu Afonso; Domenici,Fernanda Aparecida; Lataro,Renata Cristina; Portari,Guilherme Vannucchi; Vannucchi,Helio.
Changes in the metabolism of methionine can cause hyperhomocysteinemia, inducing a triad of atherosclerosis, hypertension, and increased oxidative stress. The generation of free radicals and oxidative damage to DNA is important in the liver damage caused by ethanol. In this study, the effect of methionine overload associated or otherwise with acute administration of ethanol on homocysteine values, damage to DNA, lipoperoxidation and vitamin E was evaluated. Thirty rats were divided into 3 groups: Group Ethanol 24 hours (EG24), Group Methionine 24 hours (MG24), and Group Methionine and Ethanol 24 hours (MEG24). TBARS, vitamin E, GS and, homocysteine values were determined and the Comet assay was carried out. Increased GSH, vitamin E and homocysteine levels...

Effect of nitric oxide and plant antioxidants on microsomal content of lipid radicals Biol. Res.
BOVERIS,ALEJANDRO D; GALATRO,ANDREA; PUNTARULO,SUSANA.
The antioxidant ability of nitric oxide (NO) generated by a chemical donor and of commercially available antioxidant preparations was assayed. SNAP (S-Nitroso-N-acetylpenicilamine) was used as the NO donor, and Ginkgo biloba, wheat and alfalfa preparations were tested. Lipid peroxidation was assayed by EPR employing a reaction system consisting of rat liver microsomes, ADP, FeCl3, NADPH and POBN in phosphate buffer, pH=7.4. In vitro NO exposure decreased microsomal lipid peroxidation in a dose-dependent manner. The dose responsible for inhibiting the microsomal content of lipid radical adducts by 50% (LD50) for SNAP was 550 mu M (NO generation rate 0.1 mu M/min). The addition of 50 mM hemoglobin to the incubation media prevented NO effect on lipid...

Effect of organic tomato ( Lycopersicon esculentum ) extract on the genotoxicity of doxorubicin in the Drosophila wing spot test Genet. Mol. Biol.
Dutra,Elaine S.; Dias,Cristina D.; Araújo,Bethânia C. de; Castro,Antônio J. S.; Nepomuceno,Júlio C..
The consumption of organic tomatoes (ORTs) reduces the risk of harmful effects to humans and the environment caused by exposure to toxic agrochemicals. In this study, we used the somatic mutation and recombination test (SMART) of wing spots in Drosophila melanogaster to evaluate the genotoxicity of ORT and the effect of cotreatment with ORT on the genotoxicity of Doxorubicin ® (DXR, a cancer chemotherapeutic agent) that is mediated by free radical formation. Standard (ST) cross larvae were treated chronically with solutions containing 25%, 50% or 100% of an aqueous extract of ORT, in the absence and presence of DXR (0.125 mg/mL), and the number of mutant spots on the wings of emergent flies was counted. ORT alone was not genotoxic but enhanced the...

Endothelial cell oxidative stress and signal transduction Biol. Res.
FONCEA,ROCIO; CARVAJAL,CRISTIAN; ALMARZA,CAROLINA; LEIGHTON,FEDERICO.
Endothelial dysfunction (ED) is an early event in atherosclerotic disease, preceding clinical manifestations and complications. Increased reactive oxygen species (ROS) have been implicated as important mechanisms that contribute to ED, and ROS’s may function as intracellular messengers that modulate signaling pathways. Several intracellular signal events stimulated by ROS have been defined, including the identification of two members of the mitogen activated protein kinase family (ERK1/2 and big MAP kinase, BMK1), tyrosine kinases (Src and Syk) and different isoenzymes of PKC as redox-sensitive kinases. ROS regulation of signal transduction components include the modification in the activity of transcriptional factors such as NFkB and others that result in...
